Defective thermogenesis in mPer2 mutant mice & A role of Per2 in circannual thermoregulation

چکیده
In the past few years, several molecular components of the circadian clock have been identified in various organisms. Little by little, researchers have worked out some relationships between metabolism and the circadian clock. Nowadays, due to irregular lifestyle, people suffer from a variety of disorders linked to metabolism, especially obesity, but also cancer, sleep and mental disorders. Acquiring new knowledge on peripheral clocks has become a major concern. Therefore, studying molecular regulation and circadian oscillations of tissue specific genes is of great interest. The master clock, which is located in a brain area called SCN (suprachiasmatic nuclei) links and synchronizes environmental rhythms with rhythms of peripheral clocks to optimize their metabolism. Besides integrating body signals, the central clock receives information on the surrounding environment such as altered day length or seasonal temperature fluctuations. To adapt to these changes, the circadian clock is connected to mechanisms that allow it to stay in tune with nature.

Brown fat and vascular heat dissipation
Current efforts to treat obesity and associated disorders focus on the stimulation of energy expenditure by increasing thermogenesis, for instance through activating brown adipose tissue or more recently "beige" or "brite" fat, a relatively novel type of adipose tissue with putative thermogenic potential. In this commentary, we aim to provide an alternative perspective on the current trend of a...
متن کاملDisruption of mCry2 restores circadian rhythmicity in mPer2 mutant mice.
Many biochemical, physiological, and behavioral processes display daily rhythms generated by an internal timekeeping mechanism referred to as the circadian clock. The core oscillator driving this clock is located in the ventral part of the hypothalamus, the so called suprachiasmatic nuclei (SCN). At the molecular level, this oscillator is thought to be composed of interlocking autoregulatory fe...
متن کاملPeriod2 gene mutant mice show compromised insulin-mediated endothelial nitric oxide release and altered glucose homeostasis
Period2 (Per2) is an important component of the circadian clock. Mutation of this gene is associated with vascular endothelial dysfunction and altered glucose metabolism. The aim of this study is to further characterize whole body glucose homeostasis and endothelial nitric oxide (NO) production in response to insulin in the mPer2(Brdm1) mice. We show that mPer2(Brdm1) mice exhibit compromised i...
متن کاملEthanol self-administration and reinstatement of ethanol-seeking behavior in Per1 mutant mice
Rationale Alcohol consumption shows circadian rhythmicity, i.e., alcohol preference and intake change with circadian time. Circadian rhythmicity is controlled by a biological clock, which has been shown to govern behavioral, physiological, and hormonal processes in synchronization with internal as well as external cues. Molecular components of the clock include circadian clock genes such as per...
متن کاملMPer1 and mper2 are essential for normal resetting of the circadian clock.
Mammalian Per1 and Per2 genes are involved in the mechanism of the circadian clock and are inducible by light. A light pulse can evoke a change in the onset of wheel-running activity in mice by shifting the onset of activity to earlier times (phase advance) or later times (phase delays) thereby advancing or delaying the clock (clock resetting). To assess the role of mouse Per (mPer) genes in ci...
